Fertility of Bull Semen Frozen in Skim Milk Diluent
نویسنده
چکیده
In Victoria the usual diluent for deep frozen bull semen consists of a mixture of egg yolk, sodium citrate and glycerol. The semen is equilibrated in this mixture for 17 hours prior to freezing. O’Dell and Almquist (1957), and Amman and Almquist (1957) found that survival of spermatozoa as assessed by motility was increased when the semen was diluted with a mixture of glycerol, fructose and skim milk instead of egg yolk-citrate diluent. When diluted with this mixture best survival of spermatozoa was obtained with a very short period of equilibration. The purpose of the present observations was to obtain information on the relative conception rates as distinct from spermatozoa1 motility with the two types of diluents, and to determine whether dilution with the skim milk mixture could be adapted to facilitate the processing of semen at a commercial centre, in this case the Victorian Artificial Breeders’ Co-operative, Bacchus Marsh.

Production and storage of goat semen for artificial insemination.
Environmental influences on reproduction and semen production in the buck, the problem of interaction between seminal plasma and egg yolk or milk constituents in diluent, liquid storage and processing of semen for freezing are discussed. A review is given on the use of frozen-thawed semen for artificial insemination (AI) in spontaneous and induced oestrus and factors influencing the fertility.

The feasibility of using frozen-thawed semen in caprine IVF outside the breeding season was investigated. Electroejaculated spermatozoa from a Nubian buck were washed twice and then frozen in skim milk- or in egg yolk-based extenders. Goat oocytes were matured and inseminated by frozen-thawed spermatozoa selected by swim-up. In vitro fertilization was performed in a modified defined medium (mDM...
متن کاملSperm Characteristics in Cryopreserved Buffalo Bull Semen and Field Fertility
Semen quality and its relationship to fertility are major concern in animal production. The aim of this study was to assess the relationship between frozen thawed sperm characteristics and fertility following in vivo fertility through Artificial Insemination(AI). Semen samples were collected from four buffalo bulls. Semen volume and sperm concentration appeared to be significantly different (P0...
متن کاملEffect of selection for fertility of frozen-thawed semen in chickens on the fertility of fresh and stored semen.
Studies were conducted using 68 males from the seventh generation of a line of meat-type breeder chickens selected for increased duration of fertility of frozen-thawed semen and 69 males of the randomly selected control line. These chickens were used to determine the presence or absence of differences between the selected and control lines with respect to fertility and spermatozoal motility whe...
متن کاملCryopreservation alters the levels of the bull sperm surface protein P25b.
Fertility of frozen-thawed bull sperm is reduced by cryopreservation. Freezing-thawing procedures can result in as much as a sevenfold fertility decrease. Sperm mortality and loss of motility do not fully explain the reduced fertility of cryopreserved semen; they may be partially explained by the loss of sperm surface proteins, which are necessary for fertilization. We have previously identifie...
